    What it is

    BPC-157 is a synthetic 15-amino-acid fragment related to a protective protein found in gastric juice. It is probably the most talked-about 'healing peptide' on the internet — and one of the clearest examples of the gap between online enthusiasm and published human evidence.

    The research base is real but almost entirely preclinical: rodent studies on tendon, ligament, gut and vascular injury models. Controlled human trials have not been published. FDA placed BPC-157 in Category 2 of its interim compounding list in September 2023, citing safety concerns — meaning licensed US pharmacies were told not to compound it — and its status has continued to move since, which is worth checking with a pharmacy rather than a forum.

    Tendon, ligament, muscle and gut injury models — overwhelmingly in rodents. Published controlled human trials are absent.

    How it works

    Proposed mechanisms from animal work include promoting angiogenesis (new blood-vessel growth), modulating growth-factor signaling like VEGF, and influencing nitric-oxide pathways at injury sites. 'Proposed' is doing real work in that sentence — the mechanistic map comes from rodents and cell cultures, not human studies.

    Where it came from

    Croatian researchers isolated the parent 'body protection compound' concept from gastric juice in the early 1990s, and one laboratory group has driven most of the publication record since — hundreds of rodent papers, remarkably few independent replications, and no controlled human trial in three decades. That publication pattern is itself a data point.

    • Largely one originating research group; independent replication is thin.

    What's known about risks

    FDA's 2023 Category 2 designation cited safety concerns including unknowns around immunogenicity and product quality. The deeper truth: with no controlled human data, its risk profile is simply uncharacterized — angiogenesis-promoting mechanisms also carry theoretical concerns (vessel growth is not selective about what it feeds) that nobody has studied in people.

    Where it stands legally

    Not an approved drug anywhere. Restricted from US pharmacy compounding by the 2023 action, with its category status under continued FDA review since — a moving target. What remains is the 'research use only' gray market, which is a legal posture rather than a lawful human-use channel.
