AICAR
An AMPK-activating nucleoside analog with niche clinical research history; not an approved therapeutic, and prohibited in sport.
What it is
AICAR is a nucleoside analog that activates AMPK, the cell's energy-stress sensor — which earned it the 'exercise in a pill' label after famous rodent experiments improved endurance without training. It is not a peptide, and it is not an approved drug.
It carries one of the more colorful anti-doping histories in this library: WADA banned it explicitly after evidence of use in professional cycling. Its human research history is real but narrow — older clinical work in cardiac settings — and none of it supports the athletic or fat-loss uses it's sold for.

Cardiac ischemia protection in older clinical research; endurance and metabolism in rodent studies that made it an anti-doping target.
How it works
Inside cells AICAR converts to ZMP, an AMP mimic that switches on AMPK — the master sensor that tells a cell it's low on energy. Downstream: increased glucose uptake and fat oxidation, the same program exercise activates, which is precisely why both athletes and regulators noticed it.
Where it came from
Studied since the 1980s–90s in cardiology (ischemia-reperfusion protection during bypass surgery), AICAR became famous in 2008 when rodent studies showed endurance gains without exercise. WADA added it to the prohibited list, cycling scandals referenced it, and the gray market has sold it ever since.
What the research record actually contains

What's known about risks
Its clinical-era studies involved monitored IV use in surgical patients — nothing like gray-market self-injection. AMPK activation is powerful, systemic and unstudied for chronic self-use; lactic-acid and uric-acid effects appear even in the older literature.
